What is MK-677?

MK-677 (Ibutamoren) is an orally active, non-peptide ghrelin receptor agonist (GHSR-1a) — one of the only compounds in its class that survives oral administration with meaningful bioavailability. It produces sustained GH and IGF-1 elevation through the same ghrelin receptor pathway as injectable GHSR agonists like Ipamorelin.

The oral route is the defining advantage of MK-677: all other clinically relevant GH secretagogues require injection, making MK-677 uniquely accessible for research protocols requiring oral administration or long-duration GH axis activation.

Mechanism of Action

MK-677 binds GHSR-1a in the pituitary with high affinity and selectivity, producing dose-dependent GH release. Its non-peptide structure resists gastric enzymatic degradation that destroys standard peptides, enabling meaningful oral bioavailability. The half-life of approximately 24 hours allows once-daily dosing to maintain chronic GH/IGF-1 elevation.
