What is Selank?

Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro) is a synthetic heptapeptide analogue of the immunomodulatory peptide tuftsin, developed by the Institute of Molecular Genetics of the Russian Academy of Sciences. Like Semax, Selank has been used in Russian clinical medicine — primarily for anxiety disorders and cognitive enhancement.

Selank's defining characteristic is its ability to produce robust anxiolytic effects without the sedation, tolerance, or physical dependence associated with benzodiazepines — the current standard pharmacological anxiety treatment.

Mechanism of Action

GABA-A Receptor Modulation

Selank influences GABA-A receptor function — the same receptor system targeted by benzodiazepines — but through a modulatory rather than direct agonist mechanism. This distinction is critical: benzodiazepines produce physical dependence by chronically activating GABA-A receptors, while Selank's modulatory effect appears to avoid this liability.

BDNF Expression

Similar to Semax, Selank increases BDNF expression in hippocampal and cortical tissue — suggesting long-term structural brain benefits alongside its acute anxiolytic effects.

Enkephalin Stabilization

Selank inhibits enkephalin-degrading enzymes, increasing levels of endogenous opioid peptides that contribute to mood regulation, stress response modulation, and analgesia — a novel anxiolytic mechanism distinct from GABA modulation.

Immunomodulation
