What is GHK-Cu?
GHK-Cu (Glycyl-L-Histidyl-L-Lysine-Copper(II)) is a naturally occurring tripeptide-copper complex found in human blood plasma, saliva, and urine. It was first identified by Loren Pickart in 1973 during research on how young human plasma factors influenced liver cell metabolism — and has since been the subject of over five decades of research.
What makes GHK-Cu extraordinary is the scale of its biological influence: gene expression analyses have demonstrated that GHK-Cu modulates more than 4,000 human genes — approximately 31% of the entire human genome with known function. No other peptide of its small size (340 Da) approaches this breadth of biological activity.
Mechanism of Action
Collagen Gene Activation
GHK-Cu directly upregulates expression of Collagen I, Collagen III, Elastin, and Fibronectin in fibroblasts — the structural proteins that define skin firmness, elasticity, and integrity. Simultaneously, it downregulates matrix metalloproteinases (MMP-1, MMP-2, MMP-9) that degrade existing collagen. This dual action — build more, break down less — produces measurable net increases in skin collagen density.
4,000-Gene Modulation
Among the thousands of genes GHK-Cu influences, the most relevant clusters include antioxidant defense (SOD, catalase), anti-inflammatory signaling (NF-κB inhibition), DNA repair pathways, skin barrier function, stem cell activation, and melanin regulation.
MMP Inhibition
Matrix metalloproteinases are the enzymes responsible for collagen breakdown — their activity increases dramatically with age, UV exposure, and inflammation. GHK-Cu's potent MMP inhibition is as important to net collagen retention as its synthesis stimulation.
Wound Healing
GHK-Cu has been studied as a wound healing agent for decades, consistently demonstrating accelerated wound closure, superior scar quality, and enhanced angiogenesis at repair sites.
Key Research Findings
Skin Thickness
A double-blind clinical study showed 70% increases in skin thickness measured by ultrasound following 12 weeks of topical GHK-Cu application — with significant improvements in skin laxity, fine line depth, and surface texture.
Age-Reversal Gene Signature
Perhaps the most significant recent finding: GHK-Cu treatment of aged human skin fibroblasts produces a gene expression profile that closely resembles young skin — with approximately 60% of age-related gene expression changes reversed. This 'transcriptional rejuvenation' distinguishes GHK-Cu from simple collagen stimulators.
Hair Follicle Research
Topical GHK-Cu has been shown to increase hair follicle size, shift follicles from telogen to anagen phase, and produce results comparable to established hair loss treatments in preclinical models.
Mottled Hyperpigmentation
Clinical research found GHK-Cu superior to Vitamin C for reducing mottled hyperpigmentation — suggesting mechanisms beyond antioxidant activity, likely involving direct melanocyte regulation.
